Effects of jaboticaba peel extract therapy on the prostate biology and metabolic parameters of the mice FVB in relation to aging and hyperlipidic diet

Abstract

Aging and obesity are factors which are deeply related to prostate alterations, involving the epithelium and the stroma, leading to oxidative stress, cellular proliferation and inflammatory process. The Jaboticaba (Type of black berry) shows a large variety of phenolics and anthocyanins, known to have effects in anti-inflammatory activity, hormone regulation and weight gain reduction. Thus, the main objective of the study herein is to evaluate the therapeutic effects of Jaboticaba peel extract (ECJ) on the structural and molecular biology of the prostate, considering senescence and a hyperlipidic diet, particularly related to metabolic parameters, angiogenesis, inflammation, hormone imbalance and oxidative stress in FVB mice. In addition, the effects of senescence and hyperlipidic diet will be characterized on the biology of the ventral prostate lobe. A total of 70 FVB mice will be divided into the following groups; YNG (Young), SE (senescence), SHP (senescence+hyperlipidic diet), SJI (senescence+ECJ Dose I) and SHJI (senescence+hyperlipidic diet+ECJ Dose I), SJII (senescence+ECJ Dose II) and SHJII (senescence+hyperlipidic diet+ECJ Dose II). The ECJ will be evaluated in vitro, considering the phenolic, total anthocyanin, flavonoid contents and antioxidant activity. After 60 days, all the animals will be sacrificied, and the ventral prostate lobe and liver will be collected. Metabolic parameters such as weight gain, week-long energetic consumption, insulin tolerance, fasting glucose, triglycerides, total cholesterol, cholesterol/LDL and Cholesterol/HDL will be evaluated. Histopathological analyses of the ventral prostate lobe and liver will be performed. The prostatic tissue will be evaluated by immunohistochemistry for COX-2, NF-›², CD31, VEGF, endostatin, AR and ER± antigens, in addition to microvessel density, apoptosis detection and also apoptotic index. Finally, COX-2, NF-›², VEGF, endostatin, 4HNE, GPx, GR, GSH, Catalase, SOD 1 and 2. will be evaluated by Western Blotting analyses. (AU)
