Abstract Some nuclear proteins are essential for the maintenance and regulation of the genome, transforming extracellular and intracellular signals into changes in gene expression and phenotype. In this project, we aim to analyze the nuclear proteome at different phases of the cell cycle and senescence of mouse tumor cells stimulated with FGF2. To this end, nuclear extracts will be systematically analyzed by mass spectrometry, allowing the detailed investigation of proteins closely related to the management of gene expression, cell phenotype, cell replication, chromatin remodeling and DNA repair and damage. Quantitative analysis will be performed using Spike-in SILAC and/or label free quantification. Using an integrative bioinformatics analysis, we believe will be possible to identify key molecules responsible for the induction and maintenance cell cycle arrest and senescence induced by FGF2, allowing a better understanding of cell cycle and tumor biology.
