Evaluation of the viability of Lactobacillus paracasei 431 and Bifidobacterium longum BB46 in symbiosis with acerola and amidated LM pectin citrus-based residue using two in vitro systems

Abstract

Probiotics are microorganisms that benefit human health. Prebiotics are organic compounds that the human body cannot digest. They are used as a substrate for the probiotics helping them. Some prebiotic and bioactive compounds show the capacity to boost microorganism resistance against adverse conditions to the gastrointestinal tract (GIT), allowing the probiotic to be viable by the end of colon region. The aim of this paper is to evaluate the viability of two probiotic strains; one in symbiosis with acerola residue and pectin, and another by itself, after passage through the stomach and duodenum. The chosen strains are Lactobacillus paracasei 431 and Bifidubacterium longum BB46. The acerola residue and the Amidated LM pectin citrus-based were chosen. Two in vitro systems; one in batch and another one in multistage (Microbial human ecosystem simulator - MHES) are going to be used to evaluate the viability of the strains both in isolation and in a combined way. During the simulated passage of the microorganism through the stomach and duodenum using MHES, samples are going to be extracted from the two specific regions of the simulator. These will be analyzed after cultivating them in selective media and analysis by scanning electron microscopy to see any morphology and if the viability of the bacteria was modified during its passage through the stomach and duodenum. The results are going to be analyzed by variance analysis and the individuals medium are going to be tested using the Tukey test. (p<0,05).
