Abstract

The presence of flagella in fowl non-typhoidal strains induces intestinal inflammation, reducing the systemic infection in the host. In these bacteria, the clpP gene is essential to virulence once its deletion leads to the production of excessive amount of flagella due to the inability to degrade flagellin. The fliD gene produces an essential protein for assembling the flagellum in the distal flagellar portion which facilitates the polymerization of endogenous flagellin at the edges of the growing filaments. Therefore, in the absence of the fliD gene, more flagellin monomers are exported/released to the environment. Two in vivo experiments with chickens were performed inoculating the mutant strains of Salmonella Typhimurium (Experiment 1) and Salmonella Enteritidis (Experiment 2) containing deletions of clpP and fliD genes in order to expression hyperflagellar phenotype. Comparing the infection caused by the wild-type strain, the induced hyperflagelation in the mutant strains was insufficient to abolish the ability of the pathogen to be excreted amid the feces, but on the other hand was capable of to reduce the intensity of clinical signs such as diarrhea, apathy and prostration in both experiments. Moreover, in Experiment 1 there was a significant reduction of mortality and discrete clinical signs such as bloody diarrhea and blindness. Therefore, herein we propose to perform the RNA sequencing from cecal tonsils samples harvested at three hours post-infection from young chickens infected with the chosen strains. Thus, we intend to investigate the genes and the active transcripts stimulated in infected birds at this predetermined time, clarifying the attenuation of the infection observed in the previous study.
