Abstract

Investigation of Roussoella sp. DLM-33 cultures revealed a new polyketide, roussoellatide,which encompasses an unprecedented carbon skeleton. The biosynthetic pathway proposed for roussoellatide was investigated by feeding experiments with labeled precursors. The hypothetical pathway requires at least two Favorskii-like rearrangements and an intermolecular [4+2]cycloaddition. Such reactions lack biochemical characterization. The principal aim of this proposal is to shed light on the genes encoding Favorskii oxidases and Diels-Alderases using genome mining and genetic engineering. Knock-out mutants will be generated to confirm the association of candidate genes with roussoellatide biosynthesis. A deep investigation of the unique roussoellatide biosynthesis will expand the scope of uncommon biosynthesis enzymes of fungal secondary metabolism. (AU)
