Abstract

The Alzheimer Disease (AD) is a population aging disease with great economic and social impact in public health. In the project we will developed electrochemical immunosensors for the detection of ²A1-42, P-Tau, and ADAM10 protein biomarkers in plasma and serum samples of AD patients in different stages. The detection of AD biomarkers detections requires costly and time consuming techniques. In this sense, this project proposes the development of low-cost disposable electrochemical immunosensors for AD diagnosis. Disposable electrochemical cells will be constructed by screen-printed using carbon ink. The prepared electrodes will be modified with monoclonal antibodies by layer-by-layer technique and the detection will be carry out exposing the modified electrode to a solution containing the biomarkers. After the incubation time, the modified electrodes will be exposed to a solution containing the polyclonal antibody labeled with horseradish peroxidase (HRP). The detection step will be performed electrochemically by the enzymatic reaction of HRP using hydrogen peroxide as substrate and hydroquinone as redox mediator. The detection of analytes will be carry out in fetal bovine serum fortified with different biomarkers using the developed immunosensor. After this, the immunosensor will used in real samples of AD patients at several stages. Dr. Márcia Cominetti from Department of Gerontology of Federal University of São Carlos will collaborated in the project, she has a large experience in the AD research and will support us providing real samples of AD patients.
