Abstract
This research project proposes the development of an integrated analysis system using supercritical technology and of the necessary methodology for the determination of several bioactive compounds in natural products. The proposed integrated system consists of the coupling between the sample preparation (extraction and purification) and chromatographic analysis steps. Sample preparation is accomplished through a combination of supercritical fluid extraction (SFE) or pressurized liquid extraction (PLE) with ultrasound, and the on-line coupling with the purification by solid phase extraction (SPE), which in turn is also coupled on-line with the analysis by supercritical fluid chromatography (SFC). The design of the integrated system allows performing different procedures for the extraction, purification and analysis individually (UAPLE; UASFE; SPE; SFC) or as a single online process (UAPLE/UASFE-SPE, SPE-SFC; UAPLE / UASFE-SPE-SFC). This system will be used for the development of methodologies necessary for the determination of a wide variety of bioactive compounds (phenolic acids, catechins, flavonoids, isoflavones, alkaloids, turmerones, curcuminoids, carotenoids and ecdysteroids) present in different types of samples (black tea, mate, coffee, soybeans, mushrooms, annatto seeds, turmeric rhizomes, Brazilian ginseng rhizomes and commercial extracts of these natural products). Finally an economic evaluation of the methods developed will be carried out through simulations in the software SuperPro ® Designer. (AU)
